Cancer is one of the leading causes of death worldwide. The stage at which cancer is discovered is critical for the treatment choice and overall survival of patients. Unfortunately, due to the asymptomatic nature of early-stage cancer, a lot of cancers are detected when the disease has locally spread or metastasized to other organs. Understanding the underlying mechanisms of cancer development and progression may offer clues and elements that can be used as early biomarker and/or therapeutic targets of cancer. My research is concisely summarized in the title of this thesis. The aim is to combine “mass spectrometry approaches” to enable the characterization of “proteins” from different biological systems with regard to their “structures, modifications and interactions”. But what are proteins actually? How does knowledge about structures, modifications and interactions contribute to our understanding of proteins? And why is mass spectrometry a good method to investigate proteins?..

(Nederlands: De kauwfunctie bij kinderen met cerebrale parese) De doelstellingen van dit proefschrift zijn: (1) het beschrijven van eet- en slikproblemen, inclusief kauwproblemen bij kinderen en de impact hiervan op het dagelijks leven van adolescenten en jong volwassenen met cerebrale parese (CP); (2) het ontwikkelen en valideren van een observatie instrument voor kauwen; (3) het meten van het kauwen met  kwantitatieve meetmethoden, zoals echografie, EMG en 3D kinematica om overeenkomsten en verschillen met het observatie instrument te objectiveren; en tot slot (4) het testen van de gevoeligheid van de verschillende instrumenten om verschillen in kauwbewegingen van kinderen met CP en zich normaal ontwikkelende kinderen vast te stellen. Iedereen zal het erover eens dat het gebruik van dieren in (bio)medisch onderzoek controversieel is. Het balanceren tussen  'of', 'wanneer' en 'hoe' dieren gebruikt mogen worden, blijft onderwerp van discussie. Het doel van dit proefschrift is om een bijdrage te leveren aan dit debat. The path of life of cardiac troponin T proteomic analysis of circulating proteoforms Sander Streng Troponine T is een van de meest gebruikte biomarkers voor de diagnose van een hartinfarct. Door zuurstofgebrek sterven er tijdens een infarct hartspiercellen af waardoor troponine T vanuit de cellen het bloed in komt. De diagnostische test die gebruikt wordt in een ziekenhuislaboratorium herkent het troponine T –molecuul in een bloedmonster van de patiënt. In deze studie hebben we vastgesteld dat dit molecuul van structuur verandert naar mate het langer circuleert in het bloed van de patiënt. The complement system is an important part of the innate immune system and can be divided in three different pathways; the classical, the lectin and the alternative pathway. In this thesis the role of C1q is investigated, which is the recognition molecule of the classical pathway. With the usage of epidemiological projects the clinical presentation of C1q deficient patients is investigated and the association of C1q in patients with Neuropsychiatric Systemic Lupus Erythematosus. Furthermore, using cellular based project the production and secretion of C1q by (non) immune cells is determined. Cerebral small vessel disease (CSVD) is a disease of the small perforating cerebral arteries, arterioles, venules and capillaries and is very common in the elderly population. Incidence of stroke at young age is increasing, which has been explained by an accompanying increase in the prevalence of traditional vascular risk factors. Especially for this younger age group, long-term information on the years after the stroke is of utmost importance, as these patients usually have a long life ahead. However, reliable long-term information on complications after a stroke at young age is scarce. Therefore we investigated the longterm risk of post-stroke epilepsy and its association with outcome and the long-term risk of vascular disease after a stroke at young age.
